Isolation and Culture of Neonatal Murine Primary Cardiomyocytes

چکیده
The cardiomyocyte is the main cell type in heart responsible for its contractile function. Culturing primary cardiomyocytes from mammalian sources to study their function remains challenging as they are terminally differentiated and cease multiply soon after birth. major technical hurdles associated with culture include attaining high yields, obtaining healthy/viable cells that show spontaneous contractions upon culture, avoiding contamination by non-myocyte cardiac types such fibroblasts endothelial cells. yield quality of obtained impacted a variety factors, purity reagents, composition digestion mixture, conditions, temperature tissue during different steps isolation. Here, we provide simplified workflow isolate, maintain neonatal rats/mice dishes, which can then be used study, instance, hypertrophy drug-induced cardiotoxicity. © 2021 Wiley Periodicals LLC. Isolation and culture of neonatal mouse cardiomyocytes.
Cultured neonatal cardiomyocytes have long been used to study myofibrillogenesis and myofibrillar functions. Cultured cardiomyocytes allow for easy investigation and manipulation of biochemical pathways, and their effect on the biomechanical properties of spontaneously beating cardiomyocytes. The following 2-day protocol describes the isolation and culture of neonatal mouse cardiomyocytes. We s...
متن کاملEnrichment of neonatal rat cardiomyocytes in primary culture facilitates long-term maintenance of contractility in vitro.
Long-term culture of primary neonatal rat cardiomyocytes is limited by the loss of spontaneous contractile phenotype within weeks in culture. This may be due to loss of contractile cardiomyocytes from the culture or overgrowth of the non-cardiomyocyte population. Using the mitochondria specific fluorescent dye, tetramethylrhodamine methyl ester perchlorate (TMRM), we showed that neonatal rat ca...
متن کاملIntegrin Based Isolation Enables Purification of Murine Lineage Committed Cardiomyocytes
In contrast to mature cardiomyocytes which have limited regenerative capacity, pluripotent stem cells represent a promising source for the generation of new cardiomyocytes. The tendency of pluripotent stem cells to form teratomas and the heterogeneity from various differentiation stages and cardiomyocyte cell sub-types, however, are major obstacles to overcome before this type of therapy could ...
متن کاملNon-Neuronal Acetylcholine Secretion by Neonatal Murine Cardiomyocytes
Recent studies have shown that murine cardiomyocytes possess a functional intrinsic cholinergic system that can synthesize and release acetylcholine (ACh). The way by which ACh release is regulated however, is not fully understood. Classically, ACh released from neuronal sources is regulated by the vesicular ACh transporter (VAChT). We tested the hypothesis that ACh released from neonatal murin...
متن کاملP10: Isolation and Culture of Primary Microglial Cells from Glioblastoma Patients
Glioblastoma multiform (GBM) is the most common and malignant form of glial tumors. GBM microenvironment contains various cell types showing characteristics of activated or dimorphic macrophages/microglia. Some of these cells provide significant help for tumor growth, while others are able to inhibit tumor progression. Microglia play a major role in brain function by monitoring tissue for patho...
